help: how to perform yeast lifespan assay

I use a Singer micromanipulator to do yeast lifespan assay. But the yeast cell is too small compared with the scope wire. It's difficult to move the bud from the mother cell. I wonder how to separate the cells using the micromanipulator and if there is any trick. Thank you very much.
